The C18 ProteaTips sample prep kit contains tips and reagents for desalting of peptide samples for MALDI-MS. The kit is effective in removing salts from protein digests resulting in MALDI-MS spectra with improved S/N and reduced adducts.

The SP-122 kit contains C18 ProteaTips for 1-10µL volumes, 10mL sample reconstitution and rinse solution, 10mL equilibration solution, and 10mL elution solution. The functionalized tips contain C18 beads immobilized on the inner surface of a pipette tip, maximizing the surface area in contact with the sample.

The SP-129 kit contains C18 ProteaTips for 10-200 µL volumes, 25 mL sample reconstitution and rinse solution, 25 mL equilibration solution, and 25 mL elution solution. The C18 beads are immobilized on the inner surface of a pipette tip providing a large surface area for sample capture.

BACKGROUND INFORMATION
The C18 Proteatips contain C18 reversed phase chromatography silica beads which are used for concentrating and desalting peptide samples for subsequent mass spectrometry analysis. In reversed phase chromatography, sample molecules such as proteins and peptides possessing some degree of hydrophobicity bind to hydrophobic aromatic or n-alkyl hydrocarbon (C18, C8 or C4) ligands immobilized on the silica bead matrix. Synthetic organic polymer such as polystyrene are also be used as matrix for reversed phase chromatography. The binding between the sample and the matrix occurs as a result of the hydrophobic patches of the sample molecule interacting with the hydrophobic stationary phase. The sample is bound to the stationary phase in a highly aqueous buffer and a buffer of high organic solvent content is used to dissociate the bound sample molecules. The more hydrophobic the immobilized ligand, the stronger the hydrophobic interaction and the more difficult it is to elute the sample molecules. The C18 matrices are best suited for peptides and small proteins, where as C8 for and C4 matrices are best suited for intermediate sized and large proteins respectively.

C18 ProteaTip clean-up of an in-gel protein digest:

The dual benefits of improved signal-to-noise (S/N) and increased sensitivity (144 vs. 1370 counts) synergistically assist in increasing the number of discernible peptide peaks in the MS spectrum in B.

The spectra demonstrates the effectiveness of C18 ProteaTips in desalting in-gel digested protein samples. Shown in A is a MALDI-TOF spectrum of 0.25µg, in-gel digested cytochrome C spotted directly onto a MALDI plate without desalting. Digestion was performed in 50mM ammonium bicarbonate buffer. Spectrum B is a mass spectrum of 0.25µg of the cytochrome C digest, desalted with a C18 ProteaTip. MALDI-TOF mass spectra were acquired using the ABI 4800 MALDI-TOF/TOF instrument.
